This course provides the student with a solid understanding of the fundamental concepts of High Performance Liquid Chromatography (HPLC). Designed for new operators or those wanting a refresher, this course covers an array of basic HPLC topics. Discussions include types of pumps and detectors and how they work, column selection and mechanisms of separation, solvent selection, choices in sample preparation, considerations in liquid chromatographic method development, fundamental HPLC calculations, basic HPLC system troubleshooting and an open discussion period to address any specific student needs. At the conclusion of this course, the student will have a sound basic understanding of how HPLC components and systems function, how compounds are separated by HPLC and how to make effective daily operational decisions.

I.     Introduction to High Performance Liquid Chromatography

II.     HPLC Basics

  1. What comprises a HPLC system, common terms
  2. Differences in HPLC pump designs, gradient generation and sample introduction

III.     HPLC Columns

  1. An explanation of the different modes of chromatography and how they perform separations
  2. The role of particle size and chemistry in separation mechanisms
  3. Considerations in the development of HPLC methods

IV.     HPLC Detectors

  1. A discussion of the different types of HPLC detectors, why they are chosen and how they operate
  2. The role of mobile phase in detector performance

V.     HPLC Solvents

  1. An overview of common solvents used in HPLC, their properties and tips for successful use of different solvents
  2. Common HPLC solvent modifiers and their role in the separation


VI.     HPLC Troubleshooting

  1. A summary of potential chromatographic problems and how to isolate the problem source.
  2. How to determine the source of baseline instability


VI.     Open Discussion

  1. As time allows, this time will be customized to each individual class to address specific system performance challenges. Discussions can include, but are not limited to chromatographic data solutions, advanced detectors, facility requirements, system modifications and parts or instrument sourcing.
